A group of novel 1,4-bis(3,5-dialkyl-4H-1,2,4-triazol-4-yl)bencene and 5-aryltriaz- 1-en-1-yl-1-phenyl-1H-pyrazole-4-carbonitrile derivatives have been successfully synthesized and characterized by spectroscopic analyses. The triazenes were obtained in moderate yield by a coupling reaction between 5-aminopyrazol-4-carbonitrile and aryl diazonium chlorides, and their structures were confirmed by detecting the CN functional group in both IR and C-13 NMR spectra. Conversely, the novel 1,4-bis(3,5-dialkyl-4H-1,2,4-triazol-4-yl)benzene derivatives were obtained using a previously unreported and facile pathway starting with p-phenylenediamine with selected triethyl orthoalkylates and then hydrazine monohydrate, followed by refluxing in triethyl orthoalkylates. The structure of the methyl derivative was confirmed by X-ray analysis. The synthesized compounds were tested and evaluated as antimicrobial agents.

In this work, we describe the development of the rearrangement for 7-aryl-substituted oxazolo[S,4-b]pyridines treated with aluminum chloride into synthetically hard-to-reach benzo[c][1,7]naphthyridinones. The discovered rearrangement is applied to a variety of electron-rich (hetero)arene substrates. It offers the advantages of mild conditions (90 degrees C temperature), fast reaction rates (<4 h), compatibility with air moisture, and the use of inexpensive commercial reagents. The proposed reaction mechanism and key elementary reaction acts were studied in detail using quantum chemical calculations. We previously identified a pyridomorphinan (6, SRI-22138) possessing a 4-chlorophenyl substituent at the 5′-position on the pyridine and a 3-phenylpropoxy at the 14-position of the morphinan as a mixed mu opioid receptor (MOR) agonist and delta/kappa opioid receptor (DOR/KOR) antagonist with potent antinociceptive activity and diminished tolerance and dependence in rodents. Structural variations at the 5′- and 14-positions of this molecule gave insights into the structure-activity relationships for binding and functional activity. Subtle structural changes exerted significant influence, particularly on the ability of the compounds to function as agonists at the MOR. In vivo evaluation identified compound 20 (SRI-39067) as a MOR agonist/DOR antagonist that produced systemically active potent antinociceptive activity in tail-flick assay in mice, with diminished tolerance, dependence/withdrawal, reward liability, and respiratory depression versus morphine. These results support the hypothesis that mixed MOR agonist/DOR antagonist ligands may emerge as novel opioid analgesics with reduced side effects.

Heteroarylation of alkenes with aryl iodides was efficiently achieved with a (MeDalphos)AuCl complex through Au-I/Au-III catalysis. The possibility to combine oxidative addition of aryl iodides and yr-activation of alkenes at gold is demonstrated for the first time. The reaction is robust and general (> 30 examples including internal alkenes, 5-, 6-, and 7-membered rings). It is regioselective and leads exclusively to trans addition products. The (P,N) gold complex is most efficient with electron-rich aryl substrates, which are troublesome with alternative photoredox/oxidative approaches. In addition, it provides a very unusual switch in regioselectivity from 5-exo to 6-endo cyclization between the Z and E isomers of internal alkenols.

Bacterial lipopolysaccharides (LPS) are important bio-medical structures, playing a major role in the interaction with human immune systems. Their core regions, containing multiple units ofl-glycero-d-mannoheptoses (l,d-heptose), are highly conserved structurally (withO3 andO7 glycosidic bonds), making them an epitope of high interest for the potential development of new antibiotics and vaccines. Research in this field has always been restricted by the limited availability of the parentl,d-heptose as well as its biochemical epimeric precursord-glycero-d-mannoheptose (d,d-heptose). This problem of availability has recently been solved by us, through a rapid and efficient practical synthesis ofl,d-manno-heptose peracetate demonstrated at scale. Herein we report an optimized, technically simple and versatile synthetic strategy for the differentiation of both thel-glyceroandd-glycero-d-mannoheptose scaffolds. Our approach is based on an orthoester methodology for the differentiation of all three positions of the sugar core using aO6,O7-tetraisopropyl disiloxyl (TIPDS) protecting group for the exocyclic positions. Furthermore, the regioselective opening toward 7-OH acceptors (6O-FTIPDS ethers) differentiates the exocyclic diol which has been demonstrated with a broader set of substrates and for bothmanno-heptoses for the first time.

DNA glycosylases are emerging as relevant pharmacological targets in inflammation, cancer and neurodegenerative diseases. Consequently, the search for inhibitors of these enzymes has become a very active research field. As a continuation of previous work that showed that 2-thioxanthine (2TX) is an irreversible inhibitor of zinc finger (ZnF)-containing Fpg/Nei DNA glycosylases, we designed and synthesized a mini-library of 2TX-derivatives (TXn) and evaluated their ability to inhibit Fpg/Nei enzymes. Among forty compounds, four TXn were better inhibitors than 2TX for Fpg. Unexpectedly, but very interestingly, two dithiolated derivatives more selectively and efficiently inhibit the zincless finger (ZnLF)-containing enzymes (human and mimivirus Neil1 DNA glycosylases hNeil1 and MvNei1, respectively). By combining chemistry, biochemistry, mass spectrometry, blind and flexible docking and X-ray structure analysis, we localized new TXn binding sites on Fpg/Nei enzymes. This endeavor allowed us to decipher at the atomic level the mode of action for the best TXn inhibitors on the ZnF-containing enzymes. We discovered an original inhibition mechanism for the ZnLF-containing Fpg/Nei DNA glycosylases by disulfide cyclic trimeric forms of dithiopurines. This work paves the way for the design and synthesis of a new structural class of inhibitors for selective pharmacological targeting of hNeil1 in cancer and neurodegenerative diseases.

Reported here for the first time is the Ir-catalyzed gamma-selective hydroboration of gamma-substituted allylic amides under mild reaction conditions.
