Preclinical efficacy of a gastro-sparing novel thiazolidin-4-one in alleviating secondary lesions of polyarthritis: A multi-parametric approach

The promising role of thiazolidin-4-ones (TZOs) against inflammatory conditions has been reported. From our lab, one of the TZO derivatives, compound 4C, exerted anti-inflammatory potential via inhibition of locally released cytokines and prostaglandin. In continuance, a detailed study was undertaken for the preclinical profiling of this promising TZO derivative against polyarthritis in rats, along with assessment of risk associated with the treatment. Male Sprague-Dawley rats were used for the adjuvant-induced arthritis (AIA) model. Based on the development of secondary lesion, the animals were randomized into different treatment groups. To establish the efficacy of the test compound, parameters such as inflammation, pain, disease progression, cytokines and prostaglandin (PG)-E-2 levels and complete blood cell profile were recorded along with radiological and histological examinations of joints. The study also focused on evaluating the side effect of test compound on gastric, liver, renal, blood and cardiovascular components. Compound 4C exerted promising therapeutic effect against secondary lesions in polyarthritis in rats. It limited the progression of chronic inflammation and associated pain in rats. Modulation of cytokine signalling and arachidonate metabolism by 4C was evident from inhibition of interleukin (IL)-6, tumor necrosis factor (TNF)-alpha and PGE(2) generation in AIA rats. Comparatively, compound 4C was safer than diclofenac to cause gastric, liver, renal, blood and cardiovascular toxicities. Synthesis and Antifungal Activity against Fusarium oxysporum of Some Brassinin Analogs Derived from L-tryptophan: A DFT/B3LYP Study on the Reaction Mechanism

An efficient methodology to obtain novel antifungal analogs of brassinin 1 is described. Starting from L-tryptophan 2, N, N’-dialkylthiourea 4, 4-[(1H-indol-3-yl) methylene]-2-sulfanylidene-1,3-thiazolidin-5-one 5 and alkyl (2S)-3-(1H-indol-3-yl)-2-{[(alkylsulfanyl) carbonothioyl] amino} propanoate 6 type compounds were obtained as main products in different ratios depending on the reaction conditions via a tandem dithiocarbamate formation and Michael addition reaction. In order to understand the dependence of the reaction conditions on the mechanism pathway, a DFT/B3LYP study was performed. The results suggested the existence of competitive mechanistic routes which involve the presence of an ionic dithiocarbamate intermediate 9. Antifungal activities of all products were then evaluated against Fusarium oxysporum through mycelial growth inhibition using a microscale amended-medium assay. IC50 values were thus determined for each compound. These results showed that 6-related compounds can be considered as promissory antifungal agents.

Synthesis of some 5-arylidene-2-(4-acetamidophenylimino)-thiazolidin-4-one derivatives and exploring their breast anticancer activity

Ten 2-(4-acetamidophenylimino)-5-arylidenethiazolidin-4-one derivatives 6a-k were synthesized and evaluated for their anticancer activity against MCF-7 cell line (breast adenocarcinoma). The synthetic approach involves cyclocondensation of N,N ‘-bis(4-acetamidophenyl)-thiourea (3) with ethyl bromoacetate in ethanol and sodium acetate to furnish the 2-(4-acetamidophenylimino)-4-thiazolidinone derivative 4, which underwent Knoevenagel condensation reaction with some substituted aldehydes to afford the targeted 2-(4-acetamidophenylimino)-5-arylidenethiazolidin-4-ones 6a-k. The 4-chlorobenzylidene-thiazolidin-4-one compound 6h exhibited strong inhibitory effect on the growth of breast cancer cell with IC50 (58.33 +/- 1.74 mu M), very close to that of the reference drug doxorubicin (IC50 48.06 +/- 0.36 mu M).

Flow cytometry and western blot analysis demonstrated the fact that anticancer activity was closely related with cancer cell apoptosis and the blockade of the phosphorylation of c-Met and its downstream signaling ERK and Akt. Design, synthesis, DNA assessment and molecular docking study of novel 2-(pyridin-2-ylimino)thiazolidin-4-one derivatives as potent antifungal agents

A series of novel 2-imino-4-thiazolidinone derivatives 4a,b was synthesized through reaction of unsymmetrical thioureas 3a,b with chloroacetic acid. Condensation of 4a,b with aromatic aldehydes 5a-eyielded the corresponding 5-arylidene derivatives 6a-j. In addition, the reaction of 4a,b with 4-arylazo-3-hydroxybenzaldehydes 8a-c furnished the respective mono-arylazo-4-thiazolidinones10a-f. All the newly synthesized compounds were confirmed by their elemental analysis and spectral data. The antifungal activity of the newly synthesized compounds was assessed and the compounds 6d, 6e, 6i, 6j, 9a,b and 10a-frevealedhigher antifungal activity towards Alternaria solani than to the standard Ridomil gold plus. Moreover, the DNA toxicity of 4-thiazolidinone derivatives 6d, 9a, 10b, 10c and 10f on the nucleic acid of Alternaria solani (KT354939) was performed and the results showed qualitatively more than 70% cleavage. Also, compounds 6i, 6j, 9b, 10c and 10f were docked inside the active site of 1ZOYenzyme and suitable binding with the active site of amino acids, were displayed according to their bond lengths, angles and conformational energy.

Catalytic Asymmetric Conjugate Addition and Sulfenylation of Diarylthiazolidin-2,4-diones

This work reports the first application of diarylthiazolidin-2,4-diones as nucleophiles in asymmetric catalysis. By utilizing chiral amino acid-based (thio)urea tertiary amines as the catalysts, we successively established asymmetric conjugate addition to nitroolefins and sulfenylation to N-(sulfanyl)-succinimides of diarylthiazolidin-2,4diones. Two series of biologically important 5-aryl-5-substituted thiazolidin-2,4-diones were obtained with high enantio- and diastereoselectivities (up to >99% ee and >19:1 dr). The enantio enriched adducts were found to show satisfactory anticancer activities against three different cancer cell lines using the MTT assay. All of these successes depended on the development of a general and expedient synthetic strategy to provide diverse 5H-thiazolidin-2,4-diones.

Tuning the Optoelectronic Properties of Naphtho-Dithiophene-Based A-D-A Type Small Donor Molecules for Bulk Hetero-Junction Organic Solar Cells

Four acceptor-donor-acceptor (A-D-A) type small molecules using naphtho-dithiophene as central building block, trithiophene as -bridges, malononitrile and 2-Thioxo-thiazolidin-4-one as end acceptor groups were designed by using density functional theory (DFT) and investigated as donor materials for organic solar cells (OSCs). The effects of end acceptor groups on absorption, charge transport, morphology, energy level and photovoltaic properties of the molecules were investigated and compared with reference molecule (R1= NDTP-CNCOO). These designed molecules showed relatively low HOMO levels of -5.46 to -5.56eV, strong absorption between 440-650nm by using chloroform as solvent. The designed donor molecules have an excellent electron mobility from 0.0161 to 0.0201V, hole mobility from 0.0275 to 0.0307V and open circuit voltage (Voc) from 1.76 to 1.86V. This study revealed that the designed donor materials are suitable and recommended for high performance organic solar cell devices.

A convenient and metal-free DMAP-catalyzed domino reaction of isatins, arylamines and hydroximoyl chlorides has been developed to achieve 1,3-dipolar cycloaddition of imines into aryl nitrile oxides at ambient temperature. Symmetrical aryl linked bis-iminothiazolidinones as new chemical entities for the inhibition of monoamine oxidases: Synthesis, in vitro biological evaluation and molecular modelling analysis

The multifactorial nature of Parkinson’s disease necessitates the development of new chemical entities with inherent ability to address key pathogenic processes. To this end, two series of new symmetrical 1,2- and 1,4-bis(2-aroyl/alkoylimino-5-(2-methoxy-2-oxoethylidene)-4-oxo-thiazolidin-3-yl) benzene derivatives (3a-g and 5a-e) were synthesized in good yields by the cyclization of 1,2- and 1,4-bis (N ‘-substituted thioureido) benzene intermediates with dimethyl acetylenedicarboxylate (DMAD) in methanol at ambient temperature. The bis-iminothiazolidinone compounds were investigated in vitro for their inhibition of monoamine oxidase (MAO-A & MAO-B) enzymes with the aim to identify new and distinct pharmacophores for the treatment of neurodegenerative disorders like Parkinson’s disease. Most of the designed compounds exhibited good inhibitory efficacy against monoamine oxidases. Compound 5a was identified as the most potent inhibitor of MAO-A depicting an IC50 value of 0.001 mu M, a 4-fold stronger inhibitory strength compared to standard inhibitor (clorgyline: IC50 = 0.0045 mu M). Molecular docking studies provided insights into enzyme-inhibitor interactions and a rationale for the observed inhibition towards monoamine oxidases. (C) 2016 Elsevier Inc. All rights reserved.

The IR spectra were obtained on a PerkineElmer FT-IR spectrophotometer using K.Br disks.itt NMR. and 13C NMR. of products were registered Bruker 500 spectrometer and chemical shifts are expressed as d (ppm) with tetramethylsilane (TMS) as an internal standard in.DNISO-d, as a solvent The purity of synthesized compounds was checked by chromatography and UV light (254 ran) was used for visualization. Result: A range of rhodanine and-2, 4-thiazolidinedione derivatives were synthesized using ChCll urea catalyst under solvent-free conditions at 80 C. Then, the sample derivatives were synthesized under optimum conditions Conclusion: The mild reaction conditions, good to excellent yields, easy workup, without the involxement of any harmful solvents/chemicals and easily available substrates, they all facilitated in the preparation of all synthesized compounds.
