Evaluation of new 2-hydroxy-N-(4-oxo-2-substituted phenyl-1,3- thiazolidin-3-yl)-2-phenylacetamide derivatives as potential antimycobacterial agents

A small collection of 2-hydroxy-N-(5-methyl/nonsubstituted 4-oxo-2-substituted phenyl-1,3-thiazolidin-3-yl)-2-phenylacetamides (3-16) was synthesized from the cyclocondensation of 2-hydroxy-2-phenyl-N’-[(substitutedphenyl)methylene]acetohydrazides (2) and mercaptoethanoic acid or 2-mercaptopropanoic acid, characterized with spectral and elemental analysis. In order to explore their antimycobacterial potential, newly synthesized fourteen compounds were screened for their inhibitory activity against Mycobacterium tuberculosis strain H37Rv at 6.25 mu g/mL with in-vitro primary tests. Compound 7 was found to provide the highest inhibition (98%) M. tuberculosis strain H37Rv, while most of the new derivatives showed different inhibition ratios. For the search of the putative targets which are considered as related to the antimycobacterial activity of these molecules, docking studies were performed. With molecular dynamic simulations, further possible interactions between ligands and the active site of the selected enzymes were investigated. Eventually, molecular modelling studies indicated that at least part of the mechanism of action of these compounds may be mediated by inhibition of MtInhA.

Synthesis and Reactivity of Phenylthiourea Derivatives: An Efficient Synthesis of New Thiazole-Based Heterocycles

The retentions of nonpolar solvents such as n-hexane, n-heptane, n-octane, n-nonane, n-decane and other acidic, basic and amphoteric probes such as tetrahydrofurane, dichloromethane, chloroform, acetone and ethyl acetate used without further purification on MPTT and MPMTT were measured in the temperature ranges from 303 to 333 K by inverse gas chromatography (IGC). The dispersive component of the surface energy, gamma(D)(S) of studied adsorbent surface was estimated using retention times of different nonpolar organics in the infinite dilution region. Dispersive components of the surface energies, gamma(D)(S)according to Fowkes and Dorris-Gray approaches and the acid, K-A and base, K-D constants for the 3-(6-Methyl-2-pyridyl)-2-(2-thienyl)-1,3-thiazolidin-4-one and 3-(6-Methyl-2-pyridyl)-2-(3-methyl-2-thienyl-1,3-thiazolidin-4-one were calculated.

2-(5-Chlorobenzo[d]thiazol-2-ylimino)thiazolidin-4-one derivatives as an antimicrobial agent

A series of 2-(5-chlorobenzo[d] thiazol-2-ylimino)-5-((3-(p-substituted phenyl)-1-phenyl1H- pyrazol-4-yl) methylene) thiazolidin-4-ones (3a-h) were prepared from 2-(5-chlorobenzo[d] thiazol-2-ylimino) thiazolidin-4-one (1) and 1-phenyl-3-(p-substituted phenyl)-1H-pyrazole-4-carbaldehyde (2a-h). All compounds were characterized using elemental analytical (C, H, and N) and spectral (FT-IR, H-1 NMR, C-13 NMR and GC-MS) data. These compounds were screened for their antibacterial, antifungal and antimycobacterial activities. Antimicrobial activity was evaluated against the bacterial strains e.g., Eschericha coli (MTCC 443), Pseudomonas aeruginosa (MTCC 1688), Staphylococcus aureus (MTCC 96), Streptococcus pyogenes (MTCC 442), H37Rv strain of Mycobacterium tuberculosis, and the antifungal activity was observed against strains e.g., Candida albicans (MTCC 227), Aspergillus niger (MTCC 282) and Aspergillus clavatus (MTCC 1323). The title compounds (Tz(1-10)) were synthesized by the reaction of thioglycollic acid on Schiff’s bases (L1-10) formed with 1-naphthylamine and substituted benzaldehydes. Antidiabetic effects and safety profile of chitosan delivery systems loaded with new xanthine-thiazolidine-4-one derivatives: in vivo studies

Based on important biological effects associated with xanthine and thiazolidine-4-one moieties, especially hypoglycemic and antioxidant, new xanthine derivatives with thiazolidine-4-one scaffold (XTDs, 6a-k) have been synthesized and reported by our research group, as new potential multitarget antidiabetic drugs. The goal of this work was to evaluate the in vivo antidiabetic potential and safety profile of the most proper XTDs (6c, 6e) and their chitosan based formulations, CS-XTDs (CS-6c, CS-6e) selected based on previous results in terms of their in vitro antioxidant effects and physic-chemical characteristics. Using a streptozotocin-induced diabetes model, the hypoglycemic effect (postprandial glycemia and glycated hemoglobin), biochemical markers of liver and kidney function, as well as hematological markers have been evaluated. The effect on lipid profile and clinical parameters (body weight, food, and water intake) has been also monitored. Moreover, the biopharmaceutical characteristics of 6c and 6e were predicted using in silico computerized methods. The treatment of diabetic rats with CS-XTDs, especially CS-6c was associated with appreciable hypoglycemic effect, the values of postprandial glycemia and glycated hemoglobin being similar with pioglitazone, used as reference drug. In addition, the recorded values for liver enzymes (AST, ALT, LDH), bilirubin (direct, total) and kidney biochemical markers (creatinine, uric acid, urea) were also comparable with pioglitazone. Also, CS-6c was associated with good hematological markers and clinical parameters. The results highlighted the efficiency of the developed chitosan delivery system CS-6c for the treatment of diabetes mellitus syndrome, acting as a potential multitarget agent.

An expedient synthesis of new 2-(furoxan-3-yl)thiazolidin-4-one derivatives

A series of new biologically interesting furoxan-3-thiazolidinones have been synthesized via onepot three-component reaction of furoxan aldehydes, anilines and mercaptoacetic acid. The multicomponent reaction involves condensation of furoxan aldehyde with aniline to give imine; the formed imine undergoes nucleophilic addition with mercaptoacetic acid, followed by cyclisation with loss of H2O to obtain the desired products. All the synthesized compounds were well characterized using spectral techniques and confirmed by an X-ray crystal structure for one compound.
