Concise Synthesis of Tryptanthrin Spiro Analogues with In Vitro Antitumor Activity Based on One-Pot, Three-Component 1,3-Dipolar Cycloaddition of Azomethine Ylides to Cyclopropenes

A simple, efficient and atom-economic method has been developed for the synthesis of complex alkaloid-like compounds with spiro-fused indolo[2,1-b]quinazoline and cyclopropa[a]pyrrolizine or 3-azabicyclo[3.1.0]hexane moieties. We have found that one-pot, three-component 1,3-dipolar cycloaddition reactions allow the desired products to be obtained from various cyclopropene derivatives with tryptanthrin-derived azomethine ylides generated in situ, in good to high yields and excellent diastereoselectivity. The possibility of ylide generation was exemplified by using alpha-amino acids (L-proline, L-4-thiazolidin-carboxylic acid) and simplest peptides (dipeptide Gly-Gly, tripeptide Gly-Gly-Gly). Quantum chemical investigations indicate that the reaction proceeds through the S-shaped azomethine ylide, the interaction of which with cyclopropenes proceeds via a less sterically hindered endo-transition state. The antitumor activity of some of spiro-tryptanthrin derivatives against erythroleukemia (K562), cervical carcinoma (HeLa) and colon carcinoma (CT26) cell lines was evaluated in vitro by MTS-assay.

Synthesis of Novel 2,3-Disubstituted Quinazolin-4-(3H)-ones and Their Antibacterial Activity on the Ultra-structure of Some Pathogenic Microorganisms

A SERIES of highly functionalized quinazolin-4-ones, with different substituents at position 3, have been concisely synthesized in good yields, via the reactions of 3-amino-6-bromo-2-undecyl-quinazoline-4(3H)-one with one carbon donor phenyl isothiocyante, followed by alpha-chlorinated compounds and 1,2-dichloroethanone. Moreover, reactions of 6-bromo-2-undecyl-4H-benzo-[3,1] oxazin-4-one with different hydrazides were also examined, giving new 3-substituted quinazolin-4-one derivatives. Some of the new quinazolin-4-ones were screened against gram negative and positive bacteria and showed good to moderate antibacterial activity. Structures of all new synthesized compounds in this investigation were substantiated using spectroscopic IR, H-1-NMR and MS studies.

New quinazolin-2,4-dione derivatives have been synthesized and fully characterized. The new derivatives were synthesized using 3-(2-imino-4-oxo-3H,4H,5H-thiazolidin-3-yl)-1H-quinazolin-2,4-dione by nucleophilic addition mechanism. DFT calculations using B3LYP/6-311++G(d,p) level of the theory were used to investigate the molecular structures and the relative stabilities of the anticipated isomers (E and Z). Both experimental and theoretical calculations confirmed the higher stability of the Z-isomers compared to the E-isomers. Dose-related antihyperglycemic and hypolipidemic effects of two novel thiazolidin-4-ones in a rodent model of metabolic syndrome

BackgroundThe replacement of the thiazolidinedione moiety with a thiazolidinone may yield antidiabetic compounds with similar pleiotropic effects. Hence, the aim of the present study was to explore the dose-related antihyperglycemic and hypolipidemic effects of two synthesized novel thiazolidin-4-one derivatives, one with a nicotinamide and the other with a p-chlorophenoxyacetamide substitution at the N3 position of the thiazolidinone ring (NAT1 and PAT1, respectively), in a rodent model of metabolic syndrome (MetS). MethodsMetabolic syndrome was induced in Wistar rats by neonatal administration of monosodium glutamate (i.p.) on 4 consecutive days followed by high-sucrose diet feeding for 6months. The effects of NAT1 (33 and 66mg/kg) and molar equivalent doses of PAT1 (40 and 80mg/kg) on relevant biochemical parameters were evaluated. Because MetS is a state of chronic low-grade inflammation, we also evaluated the effects of these compounds on proinflammatory markers, namely interleukin (IL)-6, tumor necrosis factor (TNF)-, reactive oxygen species (ROS), and nitric oxide (NO). ResultsBoth NAT1 and PAT1 attenuated hyperglycemia, hypertriglyceridemia, hypoalphalipoproteinemia, and glucose intolerance. PAT1 exhibited superior antihyperglycemic and antihypoalphalipoproteinemic effects than NAT1. However, NAT1 had a better triglyceride-lowering effect. At the lower dose tested, both compounds significantly reduced elevated malondialdehyde levels. In addition, PAT1 (80mg/kg) restored hepatic superoxide dismutase enzyme levels. There was a tendency for NAT1 and PAT1 to inhibit elevated hepatic IL-6 and TNF- levels, but the differences did not reach statistical significance. In addition, PAT1 exhibited in vitro anti-inflammatory activity by reducing proinflammatory ROS and NO levels in RAW264.7 macrophages. ConclusionsThe novel thiazolidin-4-ones NAT1 and PAT1 could be potential pleiotropic drug candidates targeting MetS.

Findings: QSIS assay revealed that SGK 324, SGK 327 and SGK 330 have the potentials to interfere with QS system in QS inhibitor selector1 (QSIS1) reporter strain. Moreover, these compounds were found to significantly affect QS regulated elastase and biofilm productions in Pseudomonas aeruginosa PA01 strain without affecting its growth. Molecular modeling experiments suggested that SGK 330 fits within the binding site of LasR protein of P. aeruginosa. SGK 324 and 327 also bind LasR provided that they adopt a s-trans/s-cis conformation with respect to the thiourea function. Results and Conclusion: Results of this study indicated that these compounds could be developed as new QS inhibitors without killing the bacteria but as antivirulence compounds to restrict virulence of clinically important human pathogens.
