The chelate assistance strategy was devised to promote a highly regioselective catalytic C-C bond activation reaction of saturated and unsaturated carbonyl compounds. The cationic Ru-H complex 1 was found to be an effective catalyst for mediating the coupling reaction of 1,2-disubstituted indoles with alpha,beta-unsaturated aldehydes and ketones, in which the regioselective C-alpha-C-beta activation of the carbonyl substrates has been achieved in forming the 3-alkylindole products. The analogous coupling reaction of indoles with saturated aldehydes and ketones directly led to the C-alpha- C-beta cleavage of the carbonyl substrates in forming the 3-allcylindole products. The coupling reaction of 1,2-dimethylinole with (E)-3-nonen-2-one and 2-propanol-d(8) showed 20-22% of deuterium incorporation to both alpha- and beta-CH2 of the 3-alkylindole product. The coupling reaction of 1,2-dimethylinole with (E)-3-nonen-2-one exhibited the most significant carbon kinetic isotope effect on the alpha-carbon of the product (C-alpha = 1.046). The Hammett plot constructed from the reaction of 1,2-dimethylinole with a series of para-substituted enones p-X-C6H4CH=CHCOCH3 (X = OMe, Me, H, Cl, CF3) showed a modest promotional effect by an electron-donating group (rho = -0.2 +/- 0.1). Several catalytically relevant Ru-H species were detected by NMR from a stoichiometric reaction mixture of the Ru-H complex 1 with 1,2-dimethylindole and (E)-3-nonen-2-one in CD2Cl2. These results support a mechanism of the catalytic coupling reaction via conjugate addition of indoles to enones followed by the C-C bond activation and hydrogenolysis steps.

Enones 1, are examples of available and attractive building block which already has the polyfluoroalkyl group in blocks skeleton and their functionalization is an actual and perspective approach for the synthesis of more functionalized molecules. This approach led us to new polyfluoroalkyl containing trioxabicyclo[3.3.1]nona-3,7-diene-4,8-dicarboxylates with four trifluoromethyl or two trifluoromethyl and two difluoromethyl groups in moderate yields (up to 68%). The synthesis of chiral unsaturated gamma-lactams is reported featuring a highly enantioselective palladium-catalyzed asymmetric allylic alkylation of alpha,gamma-disubstituted 2-silyloxypyrroles. This method allows a straightforward access to optically active gamma-lactams bearing an alpha-quaternary stereogenic center in high yields (up to 93%), high regioselectivities (up to >20:1), and excellent enantioselectivities (up to 95% ee). To further demonstrate the synthetic utility of the method, the resulting allylated products were converted to various versatile chiral building blocks, such as pyrrolidines and pyrrolidinones.

Shallot landraces and varieties are considered an important genetic resource for Allium breeding due to their high contents of several functional metabolites. Aiming to provide new genetic materials for the development of a novel bulb onion cultivar derived from intraspecific hybrids with useful agronomic traits from shallots, the metabolic profiles in the bulbs of 8 Indonesian shallot landraces and 7 short-day and 3 long-day bulb onion cultivars were established using LC-Q-TOF-MS/MS. Principal component analysis, partial least squares discriminant analysis, and dendrogram clustering analysis showed two major groups; group I contained all shallot landraces and group II contained all bulb onion cultivars, indicating that shallots exhibited a distinct metabolic profile in comparison with bulb onions. Variable importance in the projection and Spearman’s rank correlation indicated that free and conjugated amino acids, flavonoids (especially metabolites having flavonol aglycone), and anthocyanins, as well as organic acids, were among the top metabolite variables that were highly associated with shallot landraces. The absolute quantification of 21 amino acids using conventional HPLC analysis showed high contents in shallots rather than in bulb onions. The present study indicated that shallots reprogrammed their metabolism toward a high accumulation of amino acids and flavonoids as an adaptive mechanism in extremely hot tropical environments.

DNA glycosylases are emerging as relevant pharmacological targets in inflammation, cancer and neurodegenerative diseases. Consequently, the search for inhibitors of these enzymes has become a very active research field. As a continuation of previous work that showed that 2-thioxanthine (2TX) is an irreversible inhibitor of zinc finger (ZnF)-containing Fpg/Nei DNA glycosylases, we designed and synthesized a mini-library of 2TX-derivatives (TXn) and evaluated their ability to inhibit Fpg/Nei enzymes. Among forty compounds, four TXn were better inhibitors than 2TX for Fpg. Unexpectedly, but very interestingly, two dithiolated derivatives more selectively and efficiently inhibit the zincless finger (ZnLF)-containing enzymes (human and mimivirus Neil1 DNA glycosylases hNeil1 and MvNei1, respectively). By combining chemistry, biochemistry, mass spectrometry, blind and flexible docking and X-ray structure analysis, we localized new TXn binding sites on Fpg/Nei enzymes. This endeavor allowed us to decipher at the atomic level the mode of action for the best TXn inhibitors on the ZnF-containing enzymes. We discovered an original inhibition mechanism for the ZnLF-containing Fpg/Nei DNA glycosylases by disulfide cyclic trimeric forms of dithiopurines. This work paves the way for the design and synthesis of a new structural class of inhibitors for selective pharmacological targeting of hNeil1 in cancer and neurodegenerative diseases.

Mesoporous silica-supported Rh complex catalysts were prepared by simple silane-coupling, followed by cornplexation, and characterized by FT-IR, SEM, Rh K-edge XAFS, and elemental analysis. Local structures of the Rh complexes in each sample were almost similar to those of a nonporous silica-supported diaminorhodiurn complex. Co-immobilization of a tertiary amine on the same silica surface induced slight changes to the Rh complex structure in the case of the support with smaller pores. The prepared catalysts showed high activity for the 1,4 addition reaction of phenylboronic acids. Co-immobilization of the tertiary amine increased the reaction rate by more than 7-fold, with turnover number of nearly 8500. The catalytic performance achieved with this novel system is with much higher than that reported previously with a nonporous silica-supported catalyst. The mesoporous silica-supported Rh complex-tertiary amine showed a wide substrate scope, including unsaturated ketones and nitriles. This co-immobilized tertiary amine may activate phenylboronic acid to enhance its reactivity in the transmetalation step with Rh-OH species.

This manuscript details the impact of stabilizing agents on the structural, optical properties of Ag@SnO2 core shell nanoparticles and its biological applications. Three different stabilizing agent viz. Poly Vinyl Pyrrolidone (PVP), zeroth generation triazolyl glycodendrimer and zeroth generation triazolyl chalcone dendrimer were used as stabilizers in the synthesis of Ag@SnO2 nanoparticles. The results were compared to study their effect on the structural and optical properties of the synthesized samples. The structural parameters of the samples were investigated by XRD analysis, FESEM and HRTEM images. The presence of SnO2 shell on the Ag core was also corroborated by XPS results. The XRD data was used for refining and characterizing the elements present in the sample by Reitveld method. The average crystallite size, dislocation density, lattice parameters (a, c), lattice strain of the samples were calculated using XRD measurements. Williamson – Hall (W-H) method and Size Strain Plot (SSP) method were used to calculate the lattice strain of the synthesized sample. The concentric rings of the SAED pattern and the lattice fringe images exhibited the interplanar d-spacing values and the results were compared with XRD measurements. The optical properties of the samples were investigated using UV-Vis spectra and the band gap energy were calculated using Tauc plot. The Photoluminescence (PL) studies of the Ag@SnO2 core – shell nanoparticles with the three stabilizing agents are also discussed in this work. The antibacterial and antifungal activities of the chalcone dendrimer stabilized Ag@SnO2 core – shell nanoparticles were tested against the pathogens Bacillus subtilis, Proteus mirabilis, Candida albicans and Aspergillus niger. A number of diversely functionalized derivatives of a novel [1,3]selenazolo[4,5-d]pyrimidine have been synthesized through heterocyclization of some 2,4,5-trisubstituted-1,3-selenazoles with orthoesters in refluxing acetic acid. The synthetic compounds were evaluated for their antimicrobial activity against a panel of microorganisms including Gram-negative bacteria, Gram-positive bacteria, and pathogenic fungi. The antifungal results revealed that the new selenium-containing heterocycles were as good as or sometimes better than terbinafine and fluconazole. The in vitro anticancer activities of aforementioned heterocyclic compounds were screened against human breast carcinoma MCF-7 and HeLa cervical cancer cells as well as HDF (human dermal fibroblast) normal cells. Antiproliferative results indicated that compounds with piperidine moiety on MCF-7 cells and with morpholine moiety on HeLa cells exhibited well broad-spectrum of anticancer activities with 397, 298 and 235 mu M and 533, 390 and 204 mu M of IC(50)values after 24, 48 and 72 h of treatments, respectively, while they had no significant toxic effects on normal cells.

Some poly functionalized heterocyclic-compounds containing pyridine-moieties were readily assembled by combining differently functionalized pyridopyrimidine-6-carbonitrile derivatives1a,bwith different electrophilic and nucleophilic reagents via short synthetic routes. The structures of the prepared derivatives were ascertained from their-spectral-and elemental analyses. Some of the synthesized compounds were tested as plausible antitumor agents. Most of those tested compounds likes7,9,10,11ashowed cytotoxic potencies against different tumor cell lines. In addition, the assessments for their antioxidant activities have also been done and compound9exhibited the highest antioxidant activity while compounds7and10showed moderate activities. In an attempt to synthesise new tyrosinase inhibitors, we designed and synthesised a series of chalcone-hydroxypyridinone hybrids as potential tyrosinase inhibitors adopting strategic modifications of kojic acid. All the newly synthesised compounds were characterised by NMR and mass spectrometry. Initial screening of the target compounds demonstrated that compounds1a,1d, and1nhad relatively strong inhibitory activities against tyrosinase monophenolase, with IC(50)values of 3.07 +/- 0.85, 2.25 +/- 0.8 and 2.75 +/- 1.19 mu M, respectively. The inhibitory activity against monophenolase was 6- to 8-fold higher than that of kojic acid. Compounds1a,1d, and1nalso showed inhibition of diphenolase, with IC(50)values of 17.05 +/- 0.07, 11.70 +/- 0.03 and 19.3 +/- 0.28 mu M, respectively. The inhibition kinetics of diphenolase indicates that compounds1aand1dinduce reversible inhibition on tyrosinase. Finally, we found that copper coordination should be one of the important inhibitory mechanism of these compounds in tyrosinase.
