The synthesis of alpha-alkenyl alpha,beta-unsaturated ketones using germanium(II) salts is reported. Oxagermacycles derived from alpha,beta-unsaturated ketones with germanium(II) salts and aldehydes can be transformed into a-alkenyl a 6-unsaturated ketones. Ammonium salts promoted the elimination of Ge(II) species to afford the two classes of alpha-alkenyl alpha,beta-unsaturated ketones in good yields. The alpha-alkenyl alpha,beta-unsaturated ketones are precursors for multisubstituted heterocycles.

Recently, chiral Bronsted bases having high basicity have emerged as a powerful tool in developing new catalytic enantioselective reactions. However, such chiral strong Bronsted base catalysts are still very scarce. Herein, we report the development of a chiral anionic Brensted base having a N,N’-dialkyl ureate moiety as a basic site. Its prominent catalytic activity was demonstrated in the enantioselective addition reactions of alpha-thioacetamides as less acidic pronucleophiles with various electrophiles. Thus, the newly developed chiral catalyst with high accessibility and structural tunability would expand the scope of viable enantioselective transformations under Bronsted base catalysis.

The highly selective C = C reduction of alpha,beta-enones was realized by CF3SO3H-modifying noble-metal-free TiO2 photocatalysis. Selectivity for C = C over C = O reduction was dramatically reversed from fair (similar to 42% without CF3SO3H-modifying) to excellent (> 99%) upon only 6 mol% CF3SO3H loading without any noble-metal additive. Attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR) and C-13-nuclear magnetic resonance (NMR) demonstrated that CF3SO3H modification results in the alpha,beta-enone polar C = O bond sitting away from rather than near to the active sites of polar TiO2 catalysts.

A simple and efficient strategy for the assembly of polysubstituted thiophenes and the chemoselective reduction of alpha,beta-unsaturated ketones via the reaction of enones with elemental sulfur and by a simple switching of the reaction solvent is described. It is reported that cheap and easily available Hg(OTf)(2) can efficiently mediate the Sakurai-Hosomi reaction of N-tert-butyloxycarbonyl (Boc) amino sulfones, aldehydes, and alpha-fluoroalkyl ketones using allyltrimethylsilane, with the catalyst loading down to 0.5-5.0 mol%, enabling the facile access to synthetically valuable homoallylic alcohols or amines, respectively. A chemoselective 1,4-allylation of alpha,beta-unsaturated enones is also achieved under the catalysis of 5.0 mol% Hg(OTf)(2). In most cases, Hg(OTf)(2) exhibited intriguing properties superior to those of commonly used metal Lewis acids for such reactions, suggesting that mercury catalysis is worthwhile to explore.

Ethnopharmacological relevance: TianJiu (TJ) therapy, one type of cold moxibustion, applies to specific acupuncture points with herbal patches of hot nature, providing a constant irritant to the skin until the presence of hyperemia and blistering. Traditional and clinical reports suggest that TJ is an effective therapy for the treatment of jaundice with fresh Ranunculus sceleratus L. (RS), in which protoanemonin is one of the main irritant constituents. However, the therapeutic effect of TJ treatment with fresh RS against intrahepatic cholestasis has not been studied in animal experiments. Aim of the study: Present study was undertaken to investigate the effect of TJ treatment with fresh RS against intrahepatic cholestasis in rats and provide an experimental basis for the underlying mechanism of TJ therapy. Materials and methods: Male intrahepatic cholestatic Sprague-Dawley rats induced by 2% alpha-naphthylisothiocyanate (ANIT, 80 mg/kg B.W.) were treated by TJ therapy with fresh RS. The levels of serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), direct bilirubin (DBIL), total bilirubin (TBIL), total bile acid (TBA), hepatic malondialdehyde (MDA) and nitric monoxide (NO), as well as hepatic body ratio, bile flow and hepatic histopathological assay were measured and evaluated to investigate the therapeutic effect of TJ treatment with fresh RS. Phytochemical analysis of fresh and dried RS was performed by gas chromatography-mass spectrometer (GC-MS). Results: After TJ treatment with fresh RS, the abnormally elevated levels of serum AST, ALT, ALP, DBIL, TBIL and TBA, as well as hepatic MDA and NO at 108 h were reduced significantly (versus model group, P < 0.01). The hepatic body ratio, bile flow and hepatic pathological change of cholestatic rats at 108 h in TJ group were restored when compared with those of model group. Thirty-one compounds including lactones, flavonoids and phenolic acids were identified and determined by GC-MS analysis. The content of protoanemonin in fresh RS (9.49%) was about 25-fold higher than that in dried RS (0.38%). Conclusions: TJ treatment with fresh RS exhibited good therapeutic effect on ANIT-induced intrahepatic cholestasis in rats, which may be due to the attenuated oxidative stress in the liver tissue. It is rational for the ancients to choose fresh RS as TJ herbal patches because of its abundant protoanemonin with the character of irritant. The qualitative and quantitative results of GC-MS analysis provided the chemical basis of TJ therapy with fresh RS, which can be regarded as a simple and efficient method for the treatment of cholestasis hepatitis. Terpenes constitute one of the largest groups of natural products. They exhibit a wide range of biological activities including antioxidant, anticancer, and drug resistance modulating properties. Saffron extract and its terpene constituents have been demonstrated to be cytotoxic against various types of cancer cells, including breast, liver, lung, pancreatic, and colorectal cancer. In the present work, we have studied anticancer properties of TMPE, a newly synthesized monoterpene derivative of beta-cyclocitral-the main volatile produced by the stigmas of unripe crocuses. TMPE presented selective cytotoxic activity to doxorubicin-resistant colon cancer cells and was identified to be an effective MDR modulator in doxorubicin-resistant cancer cells. Synergy between this derivative and doxorubicin was observed. Most probably, TMPE inhibited transport activity of ABCB1 protein without affecting its expression level. Analysis of TMPE physicochemical parameters suggested it was not likely to be transported by ABCB1. Molecular modeling showed TMPE being more reactive molecule than the parental compound-beta-cyclocitral. Analysis of electrostatic potential maps of both compounds prompted us to hypothesize that reduced reactivity as well as susceptibility to electrophilic attack were related to the lower general toxicity of beta-cyclocitral. All of the above pointed to TMPE as an interesting candidate molecule for MDR reversal in cancer cells.
